5.3: Exercise #28 - Effect of Antiseptics and Disinfectants on Microbial Growth

    Exercise Outcomes:

    Course Laboratory Learning Outcomes

THECB - ACGM Lab Learning Outcome #1 Use and comply with laboratory safety rules, procedures, and universal precautions

THECB - ACGM Lab Learning Outcome #4 Perform basic microbiology procedures using aseptic techniques for transfer, isolation, and observation of commonly encountered, clinically significant bacteria.

THECB - ACGM Lab Learning Outcome #5 Use different types of bacterial culture media to grow, isolate, and identify microorganisms.

    Laboratory Exercise Learning Outcomes
    1. Define the following terms related to control of microbial growth: antiseptic, disinfectant, sterilant, sanitizer, microbicidal, microbiostatic, bactericidal, bacteriostatic.
    2. Define zone of inhibition.
    3. Complete an experiment in which a variety of chemical agents (i.e. alcohol, bleach, lysol, etc.) are tested and compared for their relative effectiveness.

    Materials Needed:

    • Bunsen burner
    • Sterile swabs
    • Broth bacterial cultures (Staphylococcus aureus, Pseudomonas aeruginosa)
    • 2 nutrient agar plates
    • Sterile disks of filter paper (1/2" diameter)
    • Microcentrifuge tubes with the antiseptic or disinfectant samples for analysis (sterile water, lysol, alcohol, bleach, hydrogen peroxide, mouthwash, hand sanitizer, pine sol, iodine, etc.)
    • Forceps
    • Beaker 70% alcohol
    • Biohazard container
