5.1: Exercise #26 - Effect of Temperature on Microbial Growth

    Exercise Outcomes:

    Course Laboratory Learning Outcomes

THECB - ACGM Lab Learning Outcome #1 Use and comply with laboratory safety rules, procedures, and universal precautions

THECB - ACGM Lab Learning Outcome #4 Perform basic microbiology procedures using aseptic techniques for transfer, isolation, and observation of commonly encountered, clinically significant bacteria.

THECB - ACGM Lab Learning Outcome #5 Use different types of bacterial culture media to grow, isolate, and identify microorganisms.

    Laboratory Exercise Learning Outcomes
    1. Define serial dilution.
    2. Define the following temperature groups used to characterize bacteria, psychrophile, psychrotroph, mesophile, thermophile.
    3. Describe how microorganisms are affected by temperature.
    4. Perform an experiment to determine temperature preferences for a given set of bacterial species, and appropriately classify the bacteria on those preferences.

    Materials Needed:

    • Bunsen burner
    • Inoculating loop
    • Broth bacterial cultures (Pseudomonas fluorescens, Serratia marcescens, Escherichia coli, Bacillus stearothermophilus)
    • 20 sterile broth cultures
